Mab1 - ab463 recognizes human, sheep, mouse, rat, porcine, rabbit and ferret HIF-1a. Bovine (-).
Mab2 - ab8366 recognizes human and rat. Mouse testing is in progress. Other species have not been tested.
Pab1 - reacts against mouse, rat, human and primate protein. Other species have not been tested.
Pab2 - Reacts with human, monkey and mouse. Other species have not been tested.

Recommended dilutions IHC-P: 1/100 - 1/500. A starting dilution of 1:1,000 is recommended with titration up or down depending upon the signal and background. IHC-Fr: 1/100 - 1/500. A starting dilution of 1:1,000 is recommended with titration up or down depending upon the signal and background.(only ab8366 has been used on frozen sections). Antigen retrieval is strongly recommended. WB: These antibodies work for Western blot analysis but are not specifically recommended for this application. Predicted molecular weight: 93 kDa.
Not tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Relevance
|
Oxygen availability plays a major role in the regulation of expression of many different genes including erythropoietin, nitric oxide synthase (NOS), heme oxygenase 1 (HO-1), glucose transporters and vascular growth factors necessary for the maintenance of homeostasis in hypoxic conditions. Hypoxia-inducible factor 1 alpha (HIF1 alpha) has been identified as a bHLH-PAS family member that is instrumental in the oxygen-dependent regulation of these genes. HIF1 alpha rapidly accumulates in the nucleus upon exposure to hypoxic conditions where it heterodimerises with the aryl hydrocarbon nuclear receptor translocator ARNT (also referred to as HIF1 beta). Heterodimerisation promotes efficient DNA binding.
