HIF1 beta (hypoxia-inducible factor 1 beta, or aryl hydrocarbon receptor nuclear transporter, ARNT) is a transcription factor that contains a basic Helix-Loop-Helix (bHLH) motif as well as two PAS domains. HIF1 beta is required for the activity of the aryl hydrocarbon (Ah) receptor, which is involved in the induction of several enzymes that are involved in xenobiotic metabolism. Binding of ligands such as dioxin to the Ah receptor result in the translocation of the Ah receptor from the cytosol to the nucleus through its association with HIF1 beta. This complex then initiates transcription of genes involved in the activation of PAH procarcinogens. Efficient DNA binding of HIF1 beta requires heterodimerisation with another bHLH protein, such as the Ah receptor.
HIF1 beta also plays a critical role in the transcriptional regulation of the adaptive response to hypoxia by forming heterodimers with HIF1 alpha and EPAS1/HIF2 alpha. Hypoxia-induced gene transcription contributes substantially to the malignant phenotype of solid tumours by promoting characteristic tumour behaviour such as angiogenesis, invasion, metastasis, de-differentiation and enhanced glycolytic metabolism.
