Blocking - HIRA peptide (ab22456)
Our Abpromise guarantee covers the use of ab22456 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
BL: Use at a concentration of 1 µg/ml.
This peptide can be used with studies using ab20655.
Not yet tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.
The HIRA gene encodes a histone chaperone that preferentially places the variant histone H3.3 in nucleosomes. Orthologs of this gene in flies, yeast and plants are necessary for the formation of transcriptionally silent heterochomatin. This gene plays an important role in the formation of the senescence-associated heterochromatin foci. These foci likely mediate the irreversible cell cycle changes that occur in senescent cells. It is considered the primary candidate gene in some haploinsufficiency syndromes such as DiGeorge syndrome, and insufficient production of the gene may disrupt normal embryonic development.
Nuclear
Blocking - HIRA peptide (ab22456)

All lanes : HIRA antibody (ab20655) at 1 ug/ml
Lane 1 : HeLa Nuclear Lysate
Lane 2 : HeLa Nuclear Lysate with HIRA peptide (ab22456) at 1 ug/ml
Lysates/proteins at 20 ug per lane.
Predicted band size : 89kDa
Observed band size : 89kDa
Additional bands at : 32kDa (possible cross reactivity).
ab20655 recognises a band at 89kDa, corresponding to the short isoform of HIRA (lane 1). This band is specifically blocked by the immunising peptide, ab22456 (lane 2). There is a band at 32kDa, which is not blocked by the immunising peptide and this is attributed to cross-reactivity.
ab22456 has not yet been referenced specifically in any publications.
