Palmisano GL et al. HLA-E surface expression is independent of the availability of HLA class I signal sequence-derived peptides in human tumor cell lines. Hum Immunol 66:1-12 (2005). Cells were treated with 500 Units of human IFN-gamma on 1 million ECV304 cells after 36 hours cells were harvested (PVG 0.1% trypsin), fixed with 2% paraformaldehyde and surface staining of HLA E was performed using ab11820 at a 1/100 dilution. The secondary used was a goat anti-mouse-FITC, at a 1/100 dilution.Red line indicates the untreated expression level and black indicate the induced surface expression of HLA E. HLA-G– or HLA-E–expressing HeLa cells were transfected with HCMV endoplasmic reticulum resident transmembrane glycoprotein US10. The lysates were immunoblotted with antibodies against HLA-G, HLA-E (ab11820), or beta-actin as a loading control. The figure shows that expression of HLA-G is slightly reduced by US10.
