Description

Mouse monoclonal [MEM-E/07] to HLA E

Specificity

The antibody recognized native surface-expressed HLA E, but not denaturated heavy chain of HLA E. The published results revealed that the antibody cross-reacts with some classical MHC Class I molecules: HLA B7 (strongly), HLA B8 (moderately), HLA B27, HLA-B44 (weakly).

Relevance

HLA E belongs to the HLA class I heavy chain paralogues. This class I molecule is a heterodimer consisting of a heavy chain and a light chain (beta-2 microglobulin). The heavy chain is anchored in the membrane. HLA E binds a restricted subset of peptides derived from the leader peptides of other class I molecules.

Cells were treated with 500 Units of human IFN-gamma on 1 million ECV304 cells after 36 hours cells were harvested (PVG 0.1% trypsin), fixed with 2% paraformaldehyde and surface staining of HLA E was performed using ab11820 at a 1/100 dilution. The secondary used was a goat anti-mouse-FITC, at a 1/100 dilution.

Red line indicates the untreated expression level and black indicate the induced surface expression of HLA E. Gating strategy - total population.

HLA-G– or HLA-E–expressing HeLa cells were transfected with HCMV endoplasmic reticulum resident transmembrane glycoprotein US10. The lysates were immunoblotted with antibodies against HLA-G, HLA-E (ab11820), or beta-actin as a loading control. The figure shows that expression of HLA-G is slightly reduced by US10.
