Recommended dilutions
ICC/IF: Use at a concentration of 1 - 5 µg/ml.
IHC-P: 1/50 for 30 min at RT
Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0 for 30 min followed by cooling at RT for 20 min.
Not yet tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.

The HMGA2 protein is a member of the High Mobility Group (HMG) family of non-histone chromatin proteins. It is expressed exclusively during mammalian embryonic development and in some cancers. HMGA2 regulates chromatin structure and gene expression by binding AT-rich sequences near promoters, altering the conformation of the surrounding DNA and thus modulating the binding of other transcription factors at the promoter. The deletion, amplification, and rearrangement of this gene are associated with myxoid liposarcoma and suggest a role in adipogenesis and mesenchymal differentiation. A gene knock out study in mouse demonstrated that this gene is involved in diet-induced obesity. The expression of HMGA2 is high in neural stem cells, and decreases with age. HMG2A promotes fetal and young adult stem cell self-renewal by decreasing p16/INK4a and p19/ARF expression levels.

Human breast carcinoma stained with
Anti-HMGA2 antibody, ab52039 at 1:50 for 30 min at RT

Immunocytochemistry/ Immunofluorescence-HMGA2 antibody(ab52039)
ICC/IF image of ab52039 stained HepG2 cells. The cells were 4% PFA fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab52039, 1µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
