For Western Blot Analysis I used your Anti-HSV1 + HSV2 gB antibody [10B7]. I had no problems with this antibody, when I worked with the wild type gB (HSV-1). But the antibody does not work with c-terminal deletion mutants of the viral protein. I would like to know which gB epitopes the antibody recognizes, but can not find any information on
your website. Is it possible to get more information about this antibody??

The immunogen used to generate the antibody clone [10B7] (ab6506) was the entire Herpes Virus. Unfortunately, the exact epitope recognised by this monoclonal antibody has not been mapped.

To our knowledge this antibody has been shown to have virus neutralizing activity, and thus it is likely that the antibody binds an extracellular domain. We unfortunately do not have data on this available though.

Maybe a search in the literature could help to find more details about this clone. The publication by Reszka and colleagues (PMID 20117270, attached) might be a good starting point.

I have finished and done two western blot experiments as well as two IF experiments, both with different concentration of antibody and got no results.

The western blot, I tried 2 times with different dilutions of antibody as well as free virus in the last blot which only showed the ladder and I've attached to this email.

I'm including the second set of IF I did and still no band just background was seen with the green channel. The second antibody 488 works well and is used often in the lab with other primary antibodies. The only thing you can see is the dapi stain.

I would really appreciate if you could send us another HSV-1 antibody that you know works well, specifically has nice images in Immunofluorescence. I think 3 weeks of trying out theAb6506antibody is enough time.
