Question 1

Tuesday 07-February-2012

Hello,I am looking for antibodies (preferentially mouseMAb so I can use same secondary), reacting withvirions of hepatitis A virus (HAV), hepatitis C virus (HCV) and parvovirus B19. I have selected ab64295 forB19 and ab8254 for HAV. Could you, please confirm these would detect the intactvirions of B19 and HAV? Also, I am not sure which of your anti-HCV antibodies to choose for this purpose. Could you, please advice?

ANSWER:

Thank you for contacting us.

Following your enquiry I have checked all our HAV, HCV and Parvovirus B19 antibodies and doublechecked with the originators of these if the antibodies would detect intact virions. Thank you for your patience while waiting for the response.

Concerning Parvovirus B19 antibody ab64295:
Indeed, we can confirm that this antibody does detect the intact virus particles. This monoclonal antibody is specific for an epitope common to VP1 and VP2 structural proteins of human Parvovirus B19. These proteins do not need to be purified or the virus does not need to be disrupted in order for the antibody to recognize the target(s).

Concerning Anti-Hepatitis A Virus antibody ab8254:
The Hepatitis A pHM-175 strain was used as the immunogen. The clone MK-01 recognises hepatitis A virus surface antigen, unfortunately no further details are available if this antibody also detects intact virions. However, I am happy to let you know that we have ab1018 available which detects intact Hepatitis A virus:

Concerning Anti-Hepatitis C Virus antibodies: As the envelope glycoproteins, E1 and E2, are essential components of the HCV virion, I have looked for our antibodies against these targets.
ab54555 and ab54556: Both antibodies can detect intact Hepatitis C virions. This has so far only been tested for the inactivated form, unfortunately not yet for non-inactivated virions.
ab20044, ab20852, ab21306 : The target protein recognized by each antibody (HCV E1 or E2) are identified by testing on Western blot. These antibodies have also been tested by ELISA and IF/IHC, but these test methods do not identify if the epitopes are recognized on intact virions. It is therefore not known if these antibodies will recognize intact HCV virions.

Please be reassured that all our products are covered by our Abpromise: We will happily offer Scientific Support in the event that a product is not functioning in the applications and species cited on the product datasheet (and the problem has been reported within 6 months of purchase). If it appears that the antibody is faulty, a replacement, credit or refund will be offered (www.abcam.com/Abpromise).

All tested applications are specified on our datasheets, which are updated as soon as any new information is brought to our attention. If you decide to go ahead and purchase one of these products, please contact our Scientific Support team prior to the purchase as you might be eligible for our testing discount program:

As we do not currently have images for all these antibodies on our datasheets, I can offer a discount off a future purchase if you buy one or more of the products now, use it a tested application and submit an image of your results to us in the form of an Abreview. The discount would be to the value of 1 free primary antibody each. The Terms and Conditions of this offer can be found at: www.abcam.com/collaborationdiscount.

Please let me know if you have any questions about this offer and I would be happy to help you further.

Question 2

Friday 30-March-2007

I would like to know if you have customer feedback on genotype 2a crossreactivity for this Ab. Thanks.

ANSWER:

Thank you for your enquiry.

Our on-line datasheets are updated as soon as any new information is brought to our attention. Unfortunately, we do not have any feedback from customers regarding genotype 2a cross-reactivity for this product.

If you require any further information, please do not hesitate to contact me.

Have a nice day.

Question 3

Friday 26-January-2007

ab20852 (Hepatitis C Virus E2 antibody [BDI167])

What is the HCV genotype/strain? I've seen BDI167 listed as genotype 1a - do you have data to support it's really 1b?

This information can be found in the datasheet:

Immunogen Recombinant full length HCV E2 protein, genotype 1b

ANSWER:

Thank you for your patience in this matter.

I have received an answer from the originator of ab20852 (Hepatitis C Virus E2 antibody). The "genotype 1a" information was an error that was discovered last year and had been corrected. The information on our present datasheet is correct:

Recombinant full length HCV E2 protein, genotype 1b

Please don't hesitate to contact us again if you have further questions.

Question 4

Monday 17-April-2006

Does this antibody cross react with genotype HCV 1b, if so how well? Where does the antibody bind on E2? What applications can be used if this antibody cross reacts with HCV 1b?

ANSWER:

Thank you for your enquiry. For ab20852, the antibody recognizes genotype 1b and the immunogen used to generate the antibody was recombinant full length HCV E2 protein, genotype 1b. The epitope has not been determined and the antibody has been tested for application in Western blot, ELISA, and Immunofluorescence.

I'm still waiting to obtain information about ab21306 and I'll get back to you when I can. Please let me know if you have any additional questions.
