Our customer is looking for  IFN gamma ELISA kit for bovine and guinea pig.   His samples are serum and plasma.   I have checked there's no product for bovine and guinea pig sample but,   please check the product which has possibility to cross react.

Thank you for your patience. I have checked on the bovine IFN-gamma ELISA Kit. Unfortunately, the kit currently is not available in our catalog as the quality control tests have not yet been completed. 

In general, due to the low homology between the human and bovine as well as guinea pig proteins, a human IFN gamma ELISA kit might most likely not work well for these two species, very unfortunately.

I also checked our IFN gamma antibodies that were tested in ELISA, but unfortunately, we do not have any that were tested in bovine or guinea pig either.

Can this TNF-a Elisa kit be used to detect TNFa in human  Plasma samples? What is the cost per kit and could we get a special evaluation price? Do you also sell sensitive kits for IFN-g? Kind regards

We have ab46025 and ab46048 to detect human IFNg in human plasma.

Hi,

We have recently purchased and run a couple of High sensitivity IFN-y ELISA kits (code ab46048). Whilst the assay worked okay and the standard curve was fine, we were a bit concerned that the OD for the top standard was only 0.4 OD units, I have run this assay before and even then the top standard OD was only ~0.7 -0.8OD units. I see in the kit insert, the example figure suggests that a top OD of ~ 2 units should be expected. Can you shed any light on why this may have occurred? We have performed the assay exactly as specified in the insert without any modifications, the only thing I can think of is the temperature of the laboratory - the insert recommends 18-25 C, though our laboratory is at the low end (~ 17- 20 C), could this relatively small change in temperature account for the large discrepancy in OD?

As an aside, on both occasions I've run this assay, we've also noticed that we haven't been provided with enough HRP diluent in the kit, it says that it should provide you with one 25ml bottle, but in actual fact only ~ 23 ml was provided.

Thanks for your enquiry. I am very sorry to hear that you are having problems with this product.

I have been discussing this enquiry with my colleagues in the Lab and would like to share the followings.

1) First, the regular volume of HRP diluent normally provided is 25.0ml and I will internally investigate how vials of only 23.0ml could have been dispatched. We can provide additional vial FOC for you if you wish. 2) Regarding low OD observed, the temperature can indeed be an explanation. The effect of lower temperature has not been fully studied and I have no data to show. However, generally based on physical-chemistry principles the temperature impacts the enzyme reactivity. In the High sensitivity kits configuration, enzyme is incubated twice (2 steps + the amplifier step) so it makes sense to consider high impact. 3) The main important factor for the good validation of the assay is the ratio between 0 and 25pg/ml standard point with 0.8 OD and less than 0.08 for zero, the assay remains fine.
