Relevance

In diploid eukaryotic cells, the chromatin fibers are about 20nM in diameter. They consist of two major components in equal amounts, DNA and basic proteins called histones. Histones are believed to be regularly arranged in the deep grove of the DNA helix. The recurring positive charges of the histones form electrostatic associations with the negatively charged phosphate groups of DNA making the DNA more stable and flexible. This allows for the supercoiling of the chromatin fibers. Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Nucleosomes consist of approximately 146 bp of DNA wrapped around a histone octamer composed of pairs of each of the four core histones (H2A, H2B, H3, and H4). The chromatin fiber is further compacted through the interaction of a linker histone, H1, with the DNA between the nucleosomes to form higher order chromatin structures. Linker histones are involved in the formation of higher order structure in chromatin and the maintenance of overall chromatin compaction. Whilst the core histones are highly conserved across a wide range of organisms, the linker histones are less conserved. Histone H2B testis specific is a variant of H2B expressed solely in the testis(SwissProt:Q96A08) and differs by the addition of three potential phosphorylation sites (serine 12, threonine 23 and threonine 34) and the repositionoing of 2 others (serine 5 and serine 6), resulting in a different phosphorylation map of the N-terminal tail.

ab23913 specifically recognises mouse Histone H2B (lane1). The signal is efficiently blocked using the immunizing peptide (lane2) but not the corresponding general human H2B peptide (lane3). This indicates that ab23913 specifically recognises testis specific H2B. To support this ab23913 does not recognise human Histone H2B in HeLa whole cell lysates (lane4).

ab23913 staining Histone H2B (testis specific) in human tesis tissue, showing a distinct and strong staining pattern within the seminiferous tubules. Paraffin embedded human testis was incubated with ab3167 (1/3000 dilution) for 30 mins at room temperature. Antigen retrieval was performed by heat induction in citrate buffer pH 6.
