Anyway, is it possible for you to supply >1mg of antibody per lot? Furthermore, what is the maximum you can supply per lot? In my eyes, I am looking for as much QC data as possible (western blot, chip, cross-reactivity dot blot, protein quantification, etc); I am not worried about your internal control QC but just what is relevant for a functional specific antibody. I would still like to receive the QC data on the lot no. that are now out of stock, if it is possible. |

Thank you for your reply. I have emailed the lab to send me all the QC data for the antibodies and lots you requested, and I will pass them on to you when I hear back from the lab. Yes, 1 mg would be possible, but it depends on the lot and antibody. For some of the lots you inquired about we might have more than 1 mg in stock, for others it would be less than a mg. This also changes daily as other customer purchase vials. But in general it would be possible, but it is lot and antibody-dependent. Hi Abcam, I can't find information about whether your antibodies (specifically Ab4441) are compatible with magnetic G beads - I understand from the Antibody information pages that your antibodies are compatible with sepharose beads, but I'm not sure if this includes magnetic sepharose beads too? I have magnetic beads XXX and would like to use them with your antibodies in a ChIP seq experiment. Will this be compatible? I'm new to this field, so thanks in advance for your help! |

Thank you very much for your inquiry. Indeed, our antibodies (as all normally purified antibodies) are in principle very well compatible with magnetic beads as well. Indeed, as long as the beads are coated to protein G and the antibody will therefore be bound by protein G, there is no problem and it will work well also if the beads are magnetic. The ab4441 is a rabbit polyclonal antibody and will therefore be bound by protein G. Chromatin was prepared from U2OS cells according to the Abcam X-ChIP protocol. Cells were fixed with formaldehyde for 10 min. The ChIP was performed with 25 µg of chromatin, 2 µg of ab4441 (blue), and 20 µl of protein A/G sepharose beads. No antibody was added to the beads control (yellow). The immunoprecipitated DNA was quantified by real time PCR (Taqman approach). Primers and probes are located in the first kb of the transcribed region.
Fruit fly (Drosophila melanogaster) Cell (polytene chromosomes) were fixed in formaldehyde, blocked in 1% BSA for 30 minutes and incubated with ab4441 (1/100) for 12 hours.
