Specificity

Current batches recognise symmetric di-methylated H4 but do not cross react with di-methylated R3 histone H2A (see image below). Previous batches have been reported to recognise both symmetric di-methylated H4 and di-methylated R3 histone H2A. This specificity was demonstrated by Ancelin et al., 2006

ChIP: Use 2-4µg for 25µg of chromatin
ICC/IF: 1/2000.
IHC-P: 1/500. Perform heat mediated antigen retrieval before commencing with IHC staining protocol.
WB: Use at an assay dependent dilution. Detects a band of approximately 16 kDa (predicted molecular weight: 11.5 kDa). Can be blocked with Histone H4 peptide - symmetric di methyl R3 (ab14791).


Not yet tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.

Relevance

Histone proteins H3, H4, H2A, and H2B function as building blocks to package eukaryotic DNA into repeating nucleosome units that are folded in higher order chromatin fibers. The nucleosome is composed of an octamer containing a H3/H4 tetramer and two H2A/H2B dimers, surrounded by approximately 146 base pairs of DNA. A diverse and elaborate array of post-translational modifications including acetylation, phosphorylation, methylation, ubiquitination, and ADP-ribosylation occurs on the N-terminal tail domains of histones. Methylation of position-specific lysine residues in histone N termini is a central modification for regulating epigenetic transitions in chromatin. Each methylatable lysine residue can exist in a mono, di, or tri methylated state. Arginine resdiues can also by mono or di methylated.

Chromatin was prepared from Hela cells according to the Abcam X-ChIP protocol. Cells were fixed with formaldehyde for 10min. The  ChIP was performed with 25µg of chromatin, 2µg of  ab5823 (blue), and 20µl of Protein A/G sepharose beads. No antibody was added to the beads control (yellow). The immunoprecipitated DNA was quantified by real time PCR (Taqman approach for active and inactive loci, Sybr green approach for heterochromatic loci). Primers and probes are located in the first kb of the transcribed region.
