    Heat shock protein of 47 kDa (Hsp 47), also called serpin H1, clade H, member 1, collagen binding protein 1 and colligin, is a member of the serpin superfamily of serine proteinase inhibitors. Hsp 47 is a 418 amino acids collagen-specific molecular chaperone involved in the collagen folding and secretion. It localizes to the endoplasmic reticulum lumen and its expression is induced by heat shock. In the sera of the rheumatic autoimmune and mixed connective tissue disease patients, Hsp 47 antigen and/or autoantibody levels were elevated. High expression of Hsp 47 was observed in ulcerative colitis-associated carcinomas cell lines and tissues. Hsp 47 is over-expressed in many fibrotic diseases such as systemic sclerosis, pulmonary fibrosis, liver cirrhosis, cicatricial pemphigoid, epidermolysis bullosa acquisita and keloids. Hsp 47 appears as a potential biomarker or therapeutic target for the treatment of collagen-related fibrosis.

    ab108861 Hsp 47 Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for detection of Human Hsp 47 in plasma, serum, tissue extract and cell culture lysates. This assay employs a quantitative sandwich enzyme immunoassay technique that measures human Hsp 47 in less than 5 hours. A polyclonal antibody specific for human Hsp 47 has been pre-coated onto a 96-well microplate with removable strips. Hsp 47 in standards and samples is sandwiched by the immobilized antibody and biotinylated polyclonal antibody specific for Hsp 47, which is recognized by a streptavidin-peroxidase conjugate. All unbound material is then washed away and a peroxidase enzyme substrate is added. The color development is stopped and the intensity of the color is measured.
