Human TRAIL (CD253) in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit (ab46074) is designed for the quantitative measurement of TRAIL in cell culture supernatants, buffered solutions or Human serum, plasma, and other body fluids.
A monoclonal antibody specific for TRAIL has been coated onto the wells of the microtiter strips provided. Samples, including standards of known TRAIL concentrations, control specimens or unknowns are pipetted into these wells. During the first incubation, the standards or samples and a biotinylated monoclonal antibody specific for TRAIL are simultaneously incubated. After washing, the enzyme Streptavidin-HRP, that binds the biotinylated antibody is added, incubated and washed. A TMB substrate solution is added which acts on the bound enzyme to induce a colored reaction product. The intensity of this colored product is directly proportional to the concentration of TRAIL present in the samples.
This kit will recognize both endogenous and recombinant Human TRAIL.

Relevance
TRAIL is a cytokine that belongs to the tumor necrosis factor (TNF) ligand family. It preferentially induces apoptosis in transformed and tumor cells, but does not appear to kill normal cells although it is expressed at a significant level in most normal tissues. TRAIL binds to several members of TNF receptor superfamily including TNFRSF10A/TRAILR1, TNFRSF10B/TRAILR2, TNFRSF10C/TRAILR3, TNFRSF10D/TRAILR4, and possibly also to TNFRSF11B/OPG. The activity of TRAIL may be modulated by binding to the decoy receptors TNFRSF10C/TRAILR3, TNFRSF10D/TRAILR4, and TNFRSF11B/OPG that cannot induce apoptosis. The binding of TRAIL to its receptors has been shown to trigger the activation of MAPK8/JNK, caspase 8, and caspase 3. -
