Question 1

Tuesday 10-April-2012

Thank you for your email.
Then, ab67943 detects only ICAM4? Our customer wants to check this.
Best regards,

ANSWER:

Thank you for your e-mail.

Currently, we don't have cross-reactivity data for ab67943. The immunogen used to raise this antibody is from ICAM4 (NP_001034221.1, 1 a.a. ˜ 272 a.a) full-length human protein.

This is a polyclonal antibody and for polyclonals (which recognize multiple epitopes) we don't do epitope mapping. Thus we can't provide you the exact epitope sequence where the antibody is targeting on.

As a general guidance, if the sequence similarity between immunogen and its isoforms (or other proteins you are interested in) is higher than 95%, it indicates that the antibody may cross-react with other family members.

I hope this will be useful for you and your customers.

Question 2

Wednesday 04-April-2012

Dear Technical team,

Our user ordered ab67943. She would liketo check someinformation.
She knows that ICAM4has sequence homology with ICAM 1,2 and 3. But, She could find detail information on your data sheet.
So, if you have information about homology and epitope.
Please let me know.

Best regards,

ANSWER:

Thank you for your inquiry.

As the datasheet indicates, the immunogen used to raise this antibody was a full length protein, corresponding to amino acids 1-272 of Human ICAM4.

This antibody is a mouse polyclonal antibody. Polyclonal antibodies consist of a mixed population of IgGs each of which will recognize a different epitope.

I hope this information is helpful to you. Please do not hesitate to contact us if you need any additional information or advice.

Question 3

Friday 20-January-2012

I have questions about whether these two abs (Anti-ICAM4 antibody (ab67943) and Anti-ICAM4 antibody (ab112554)) are an intact IgG containing the Fc region? If not, would you help us to find one lacking the Fc region? Or if is it possible to custom-order these abs lacking the Fc from your company?

ANSWER:

Thank you for contacting Abcam.

Both ab67943 and ab112554 are intact IgG molecules containing the Fc region. I have looked through our catalogue and I cannot find anything that matches your requirements, but you could try searching http://www.biocompare.com.

Question 4

Monday 26-September-2011

Thanks for your reply. I have 3 additional antibodies that I would like to test in flow cytometry and obtain a discount code for an Abreview.

Anti-CD135: ab37847

Anti-EDIL3 ab74775

Anti-CD105 ab21224

ANSWER:

I am very pleased to hear you would like to test these 3 antibodies in flow. These codes will give you 1 free primary antibody each before the expiration date. To redeem this offer, please submit an Abreview for flow and include this code in the “Additional Comments” section so we know the Abreview is for this promotion. Remember, we publish both positive and negative Abreviews on our datasheets so please submit the results of your tests. The code will be active once the Abreview has been submitted and can be redeemed in one of the following ways: 1) Call to place your order and mention the code to our customer service department; 2) Include the code in your fax order; 3) Place your order on the web and enter the promotional code.

Any feedback that you can provide will be greatly appreciated, whether positive or negative. If you have any further questions, please do not hesitate to contact us. We look forward to receiving your Abreview and wish you luck with your research.

Question 5

Monday 19-September-2011

Dear Jeremy-

In our conversation yesterday I had written down that we decided on the anti-ICAM4 antibody ab112554...because it had been tested via ELISA and the immunogen was a large extracellular portion of the molecule....? the offer below is for ab67943, an antibody that is not certified for ELISA?

In addition to ab112554, I would like to test 3 more of your polyclonal antibodies using flow cytometry: human EDIL3, human endoglin (CD105), and human FLT3 (CD135). I have a donkey-anti rabbit APC secondary antibody so I would like to stick with rabbit anti-human for all 4 antibodies.

My experiments involve using these antibodies to (1) see if these surface proteins are expressed on my target cells using flow cytometry (2) use these antibodies to block these surface proteins. Part2 will work better if I use a polyclonal not monoclonal antibody.

How can I determine that the immunogen for each polyclonal antibody includes the extracellular domain? Should I pick those generated against the largest extracellular domain immunogen if I want to achieve the best blocking ?? Is this information available on the website or do I need to get that information from you??

Thanks for you help,

ANSWER:

Thanks for your email. Sorry for the confusion with the antibody I offered incorrectly for the promotion. Below please find a new discount code/expiration date for testing ab112554 in flow.

Regarding your questions:

1. How can I determine that the immunogen for each polyclonal antibody includes the extracellular domain?

===>Typically, if available, the immunogen information for an antibody will be noted in the 'Immunogen' section of the online datasheet. If you are looking for more information, please contact our technical support team as we are often able to provide more details.

2. Should I pick those generated against the largest extracellular domain immunogen if I want to achieve the best blocking?

===>Unless 'Blocking' (Bl) or 'Neutraliztion' (Neut) is noted in as a tested application, we do not know if the antibody will work to block the receptor/ligand binding. However in general, I agree with you in that you're best chance of success with those applications is to test a polyclonal raised against the entire extracellular domain

I hope this is helpful. Please contact me if you need more help choosing an antibody for your experiments.
