I am sorry I am not able to confirm the expression level of IGF1 in skeletal protein with a literature search. Do you have any informationon this? I can suggest to consider including a positive control sample such as plasma, A549 cell line lysate orin this case. 2. Could you confirmif there is there any background staining on the blot? 3. I can suggest to consider try RIPA lysis buffer which may provide a more suitable protein preparation for this protein and epitope. 4. Could you confirm ifthe samples heated to reduce and denature? Please provide details. I can also recommend it would be beneficial to use a reducing gel. We suggest to use reducing and denaturing conditions for western blot with our antibodies, unless it recommends otherwise on the datasheet. 5. I can suggest to try BSA rather than fish gelatin to block. Changing blocking agents can sometimes improve the results. 6. I recommend to try the antibody at a higher concentration in order to increase the signal. Try 1:1000 and 1:500. 7. Is the secondary antibody working well with other primary antibodies?
