Hello,
I would like to test the Anti-IGF1 antibody (ab40657) in IHC ofparaffin-embedded mouse prostate tissue. According to the datasheet, this antibody recognizes human IGF1, but it hasn't been tested on mouse.
Could you let me know if I would get the 100% testing discount if I do the test and submit the abreview?
Also, I welcome any suggestions for the test that might not be included in the datasheet. Or suggestions of other antibodies that might be more suitable for my needs.

Dear technical team,   Our customer would like to purchase ab40657 and has inquiry about immnogen information. http://www.abcam.com/IGF1-antibody-ab40657.html   It described as "Synthetic human insulin-like growth factor 1 [Ac60-70] covalently attached onto a carrier protein."   1. Why is this attached onto a carrier protein?   The customer asked about the role of carrier protein and effect on antibody activity.   2.  What is the meaning of "Ac" ([Ac60-70] )?   Check them please.   Thank you.

Thank you for your enquiry and your interest in our products.

Native human IGF-1 is a 70 amino acid long peptide. We wanted to develop an antibody that bounds specifically to the -COOH terminus of the whole peptide, so we synthesized an 11-mer, i.e. residues #60-70 aa - to use as immunogen.

Since we didn't want a free NH2-group at the amino terminal, we capped the amino terminus during synthesis by acetylating it with acetic anhydride which adds an acetyl group, i.e. the Ac moiety. Thus the resulting synthetic analogue of the carboxyl terminal region is Ac60-70.

This synthetic peptide was coupled onto bovine thyroglobulin as carrier protein to elicit an antibody response since the immune system will not recognize small peptides, such as this 11-amino acid long analogue, as an antigen without it being conjugated onto a much larger protein.

We received a lyophilised antibody. data sheet said dilute with 0.1 ml 0f 10mg/mlBSA in PBS. Please simplify. we have only PBS of distilled water as diluent. how many ml do we have to add.please answer asap. thanks.

I am sorry to hear that you are having problems with using one of our products.

This product is supplied as lyophilized powder, which should be stored at 4oC until rehydrated. Histochemical grade antibodies should be diluted with PBS that contains either 1.0% BSA or 1% normal goat serum, aliquoted into single use portions, and stored frozen at -20C: do not repeatedly freeze and thaw.
