    Interleukin-1 alpha (IL1 alpha) is a member of the IL1 superfamily containing ILl alpha, ILl beta, and IL1Ra receptor antagonist. IL1 alpha is known as hematopoietin (IL1F1) and ILl beta as catabolin (IL1F2). IL1 alpha and ILl beta are corresponding to two different isoelectric forms, acidic pI 5 and neutral pI 7, respectively. IL1 alpha has a molecular mass of 17 kDa and consist of 159 amino acids having 26-33% homology with ILl beta. They are produced mainly by macrophages and monocytes, processed and released during apoptosis, and bind with high affinity to specific receptors on target cells. While only the mature form of ILl beta has biological activity, both the pro and mature forms of IL1 alpha are active. IL1 alpha and beta are pro-inflammatory cytokine involved in immune responses, inflammatory reactions, and hematopoiesis. IL1 alpha is an epidemal cytokine that is constitutively produced by epithelial cells and plays important role in maintenance of skin barrier function. The polymorphism of increased IL1 production in patients is associated with rheumatoid arthritis, coagulation, solid tumors, leukemias, Alzheimer’s disease, autoimmune disorders, and myocardial infarction.

    ab108864 IL1 alpha Human ELISA kit is designed for detection of human IL1 alpha in plasma, serum, and cell culture supernatants. This assay employs a quantitative sandwich enzyme immunoassay technique, which measures IL1 alpha in 5 hours. A murine monoclonal antibody specific for IL1 alpha has been pre-coated onto a microplate. IL1 alpha in standards and samples is sandwiched by the immobilized antibody and a biotinylated polyclonal antibody specific for IL1 alpha, which is recognized by a streptavidin-peroxidase conjugate. All unbound material is then washed away and a peroxidase enzyme substrate is added. The color development is stopped and the intensity of the color is measured.

  • RelevanceIL1 alpha is a member of the interleukin 1 cytokine family. It is a pleiotropic cytokine involved in various immune responses, inflammatory processes, and hematopoiesis. IL1 alpha is produced by monocytes and macrophages as a proprotein, which is proteolytically processed and released in response to cell injury, and thus induces apoptosis. This gene and eight other interleukin 1 family genes form a cytokine gene cluster on chromosome 2. It has been suggested that the polymorphism of these genes is associated with rheumatoid arthritis and Alzheimer's disease.
