Protein descriptionRecombinant full length mature Human IL1 alpha (amino acids 113-271) with His tag produced in the endosperm tissue of barley grain (Hordeum vulgare); 175 amino acids with tag, Predicted MWt 20.2 kDa (UniProt P01583).
Expression hostBarley
Properties
Purity> 95
% by SDS-PAGE
Purification notesSterile filtered through a 0.2 µm filter.
Endotoxin level is less than 0.005ng per µg (0.05EU/µg) as measured by turbidimetric kinetic assay.
ab88276 carries no pyrogenic or pro-inflammatory contaminants, as assayed with monocyte activation test using Human 10-plex Cytokine Assay measuring IL6, TNF-alpha and IL1beta induction.
Biological activityBioactivity:
The bioactivity is determined by the cell proliferation assay for IL1 alpha for dose-dependent cell proliferation of the interleukin 1 (IL1) sensitive murine T-helper cell line D10S, a stable line which proliferates to subfemtomolar concentrations of IL1 in the absence of mitogens. The effect of serial dilutions of IL1 alpha on the proliferation of D10S cells is monitored at OD490 nm. The ED50 lies in the range of 2.2-3.3 pg/ml corresponding to 4x 108 U/mg.
FormLyophilised:Reconstitute the lyophilized protein in sterile water to a concentration of no less than 100 µg/ml. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).
Storage instructionsThe lyophilized protein is stable for a few weeks at room temperature, but best stored at -20°C.This product is an active protein and may elicit a biological response in vivo, handle with caution.
Storage bufferPreservative: None
Constituents: PBS, pH 7.2

Additional notesReconstituted protein should be used immediately or stored in working aliquots at -20°C. Avoid repeated freeze-thaw cycles.

FunctionProduced by activated macrophages, IL-1 stimulates thymocyte proliferation by inducing IL-2 release, B-cell maturation and proliferation, and fibroblast growth factor activity. IL-1 proteins are involved in the inflammatory response, being identified as endogenous pyrogens, and are reported to stimulate the release of prostaglandin and collagenase from synovial cells.
Sequence similaritiesBelongs to the IL-1 family.
DomainThe similarity among the IL-1 precursors suggests that the amino ends of these proteins serve some as yet undefined function.
Cellular localizationSecreted. The lack of a specific hydrophobic segment in the precursor sequence suggests that IL-1 is released by damaged cells or is secreted by a mechanism differing from that used for other secretory proteins.
