    Molecular Mass: ab89013 is a disulfide-linked heterodimeric glycoprotein, consisting of an alpha and beta chain. ab89013 migrates as multiple bands between 30 and 45 kDa in SDS-PAGE due to post-translational modifications, in particular glycosylation. ab89013 IL12 alpha chain migrates as bands between 30 and 35 kDa. This compares with the unmodified IL12 alpha chain that has a predicted molecular mass of 22.5 kDa. ab89013 beta chain migrates as bands between 35 and 45 kDa. This compares with the unmodified IL12 beta chain that has a predicted molecular mass of 34.7 kDa. ab89013 IL12 alpha chain consists of 15-35% carbohydrate by weight. ab89013 IL12 beta chain consists of 0-25% carbohydrate by weight.

    pI: ab89013 separates into a number of isoforms with a pI between 3.5 and 9.5 in 2D PAGE due to post-translational modifications, in particular glycosylation. ab89013 IL2 alpha chain separates into multiple isoforms with a pI between 3.5 and 9.5. This compares with the unmodified IL12 alpha chain that has a predicted pI of 6.1. ab89013 IL12 beta chain separates into multiple isoforms with a pI between 5.0 and 9.0. This compares with the unmodified IL12 beta chain that has a predicted pI of 5.4.

    • 1D SDS-PAGE of ab89013 before and after treatment with glycosidases to remove oligosaccharides.
      Lane 1: ab89013
      Lane 2: ab89013 treated with PNGase F to remove potential N-linked glycans
      Lane 3: ab89013 treated with a glycosidase cocktail to remove potential N- and O-linked glycans.

      Appearance of additional band at lower MWt after treatment with PNGase F indicates the presence of N-linked glycans. The subsequent drop in MWt after treatment with a glycosidase cocktail indicates the presence of O-linked glycans. Additional high MWt bands in lane 3 are glycosidase enzymes.
    • A sample of ab89013 without carrier protein was reduced and alkylated and focused on a 3-10 IPG strip then run on a 4-20% Tris-HCl 2D gel. 40 µg of protein was loaded. Spot trains indicate presence of multiple isoforms.
    • Densitometry of protein isoforms visualised by 2-DE.
      The densitometry scan demonstrates the purified human cell expressed protein exists in multiple isoforms, which differ according to their level of post-translational modification.
      The triangles indicate theoretical pI and MWt of the alpha and beta chains of the protein.
