Protein descriptionRecombinant full length mature Human IL4 (amino acids 25-153) with His tag produced in the endosperm tissue of barley grain (Hordeum vulgare); 146 amino acids with tag, Predicted MWt 17.2 kDa (UniProt P05112).
Expression hostBarley
Properties
Purity> 95
% by SDS-PAGE
Purification notesSterile filtered through a 0.2 µm filter.
Endotoxin level is less than 0.005ng per µg (0.05EU/µg) as measured by turbidimetric kinetic assay.
ab89578 carries no pyrogenic or pro-inflammatory contaminants, as assayed with monocyte activation test using Human 10-plex Cytokine Assay measuring IL-6, TNF-alpha and IL-1beta induction.
Biological activityThe bioactivity is determined by the cell proliferation assay for IL4 for dose-dependent cell proliferation of Human TF-1 cells. The ED50 for ab89578 lies in the range of 0.8-1.2 ng/ml corresponding to 1.2-0.8x106 U/mg.
FormLyophilised:Reconstitute the lyophilized protein in sterile low salt buffer to a concentration of no less than 100 µg/ml. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).
Storage instructionsThe lyophilized protein is stable for a few weeks at room temperature, but best stored at -20°C.This product is an active protein and may elicit a biological response in vivo, handle with caution.
Storage bufferPreservative: None
Constituents: 0.7M Sodium chloride, 30mM Sodium acetate, pH 5.3

Additional notesReconstituted protein should be used immediately or stored in working aliquots at -20°C. Avoid repeated freeze-thaw cycles.

FunctionParticipates in at least several B-cell activation processes as well as of other cell types. It is a costimulator of DNA-synthesis. It induces the expression of class II MHC molecules on resting B-cells. It enhances both secretion and cell surface expression of IgE and IgG1. It also regulates the expression of the low affinity Fc receptor for IgE (CD23) on both lymphocytes and monocytes.
Involvement in diseaseGenetic variations in IL4 may be a cause of susceptibility to ischemic stroke (ISCHSTR) [MIM:601367]; also known as cerebrovascular accident or cerebral infarction. A stroke is an acute neurologic event leading to death of neural tissue of the brain and resulting in loss of motor, sensory and/or cognitive function. Ischemic strokes, resulting from vascular occlusion, is considered to be a highly complex disease consisting of a group of heterogeneous disorders with multiple genetic and environmental risk factors.
Sequence similaritiesBelongs to the IL-4/IL-13 family.
Cellular localizationSecreted.
