This will help my understanding of the assay and how it is working with these antibodies. 1. Was the recombinant protein full length influenza A virus Hemagglutinin H1 protein? Does it contain the immunogen sequence for these antibodies? 2. How was it prepared? 3. How much HA recombinant protein(in ug) was added to each well? Has this been optimised with the dilution of antibody? It may be there is too much HA recombinant protein for the amount of antibody to totally neutralizse. 3. Here is an answer from our customer: I will be happy to present all necessary information for the troubleshooting process. "It looks like ab66189 and ab61986 are being use as detection antibodies which are in turn detected by anti-mouse secondary antibodies? Is this the case?" Yes, we used these antibodies as detection antibodies which are in turn detected by anti-mouse secondary antibodies (biotin- labeled secondary antibody, Sigma) "If so I was wondering how much of these antibodies are being added to the wells (eg. 1ug/well) and if any other dilutions have been tried?" We added to the wells only 1.01ug/well of ab66189 and 0.64ug/well of ab61986. Hope that will be helpful.

It does appear as if these two antibodies are not working as expected in ELISA. I would be happy to replace them for you with any other primary antibodies.
