Performed under reducing conditions.
Protocol notes:
- SDS-PAGE in reducing conditions, 10 % acrylamide
- After SDS-PAGE transfer to nitrocellulose membrane
- Blocking in PBS, containing 0.5% Tween 20, 10% dry milk (w/v) – incubation 10 minutes at room temperature.
- Primary antibody: 5 µg/ml diluted in PBS containing 0.5% Tween 20 – incubation 1 hour at room temperature.
- Secondary antibody: HRP-conjugated anti-mouse IgG diluted in PBS, containing 0.5% Tween 20 – incubation 1hour at room temperature.
- Diaminobenzidine 50 µg/ml – 20 min
