I will be happy to present all necessary information for the troubleshooting process.

"It looks like ab66189 and ab61986 are being use as detection antibodies which are in turn detected by anti-mouse secondary antibodies? Is this the case?"

Yes, we used these antibodies as detection antibodies which are in turn detected by anti-mouse secondary antibodies (biotin- labeled secondary antibody, Sigma)

"If so I was wondering how much of these antibodies are being added to the wells (eg. 1ug/well) and if any other dilutions have been tried?"

We added to the wells only 1.01ug/well of ab66189 and 0.64ug/well of ab61986.

I'm looking for an H1N1 antibody that cross-reacts with Mexico/2009 strain.

ANSWER:

 

I would still need the accession number for your strain, but I just wanted to let you know of additional information about 2 of our products.


Concerning ab61986, this antibody recognizes all Hemagglutinin from H1N1 strains. The immunogen used to produce this antibody is from Influenza A/USSR/90/77. The epitope is in the range aa206-225 (based on accesssion # ABD95350).



For ab66189, this antibody has not been tested for reactivity to Influenza A/Mexico/2009 strain. The immunogen used to develop ab66189was purified Influenza A/New Caledonia/20/99 (H1N1) (not a synthetic peptide sequence). This clone has been tested to show reactivity to hemagglutinin H1, but the exact epitope location on H1 has not been mapped. For this reason, we are not able
to do a BLAST to compare to A/Mexico/2009.



If you can provide the sequence, Icanget the BLAST done on ab91531.
