Influenza A virus is a major public health threat. Novel influenza virus strains caused by genetic drift and viral recombination emerge periodically to which humans have little or no immunity, resulting in devastating pandemics. Influenza A can exist in a variety of animals; however it is in birds that all subtypes can be found. These subtypes are classified based on the combination of the virus coat glycoproteins hemagglutinin (HA) and neuraminidase (NA) subtypes. In natural infection, inactive HA is matured into HA1 and HA2 outside the cell by one or more trypsin-like, arginine-specific endoprotease secreted by the bronchial epithelial cells.

  • SDS-PAGE in reducing conditions, 10 % acrylamide
  • After SDS-PAGE transfer to nitrocellulose membrane
  • Blocking in PBS, containing 0.5% Tween 20, 10% dry milk (w/v) – incubation 10 minutes at room temperature.
  • Primary antibody: 5 µg/ml diluted in PBS containing 0.5% Tween 20 – incubation 1 hour at room temperature.
  • Secondary antibody: HRP-conjugated anti-mouse IgG diluted in PBS, containing 0.5% Tween 20 – incubation 1hour at room temperature.
  • Diaminobenzidine 50 µg/ml – 20 min
