I have got a question about your antibody Anti-Integrin beta 3 antibody [EP2417Y] (ab75872). Is it against isoform alpha-V or isoform beta-3 or is it combined against isoform alpha-V and isoform beta-3? If it´s not combined, do you have a combined one? |

I have confirmed that the antibody ab75872 is raised against the C-terminus of isoform beta 3a. The immunogen is within the range aa 763 - 788. If you have the amino acid sequence of isoform alpha-V, I will be happy to align it with the beta 3a range, which will allow us to assess the potential for cross-reactivity with alpha V. To our knowledge, there is no data that demonstrates cross-reactivity with other isoforms. |

I will need to ask the developer of this clone to confirm but it appears to be raised against the C terminus of integrin beta 3, isoform beta a, based on the specificity note on the datasheet: "ab75872 detects Integrin beta 3A. It does not detect Integrin beta 3B or Integrin beta 3C". Here is a link to the UniProt datasbase entry for integrin beta 3: http://www.uniprot.org/uniprot/P05106 Do you have the amino acid sequence for isoform alpha V? To assess potential cross-reactivity of the antibody with this isoform, we will need to align the isoform sequence with that of the immunogen. Do you mean integrin alpha V? ab75872 staining mouse brain sections by IHC-P. The tissue was fixed with paraformaldehyde and a heat mediated antigen retrival step was performed with citric acid pH 6. Permeabilization was performed with 0.1% Tween and blocking of the sample was done with PBS/0.1% Tween/5% goat serum for 1 hour at 25°C, followed by staining with ab75872 at 1/250 in PBS/0.1% Tween/5% goat serum/azide for 12h at 4°C. A biotinylated goat anti-rabbit polyclonal antibody at 1/200 was used as the secondary antibody. Upper panel shows a section from a WT animal and bottom panel shows a section from an animal with full knockout to the Itgb3 gene. Integrin beta3 staining was observed in NeuN positive cells.

ab75872 staining Integrin beta 3 in Human pancreatic PanIN lesion tissue by Immunohistochemistry (Frozen sections). The sections were fixed in Acetone prior to blocking with 5% Serum for 1 hour at room temperature. The primary antibody was diluted 1/100 in PBS and incubated with the sample for 8 hours at 4ºC. An Alexa Fluor® 488-conjugated Goat anti-Rabbit polyclonal was used as the secondary antibody, diluted 1/1000.
