Application notes

ICC/IF: Use at a concentration of 1 µg/ml. ab2283 clearly stains the cytoplasm as expected. Foci within the nucleus are also seen. We have no explanation for this and we would appreciate any customer feedback.

WB: Use at an assay dependent dilution. Predicted molecular weight: 50 kDa. Can be blocked with Integrin linked ILK peptide (ab13748).


Not yet tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.

Target

Function

Receptor-proximal protein kinase regulating integrin-mediated signal transduction. May act as a mediator of inside-out integrin signaling. Focal adhesion protein part of the complex ILK-PINCH. This complex is considered to be one of the convergence points of integrin- and growth factor-signaling pathway. Could be implicated in mediating cell architecture, adhesion to integrin substrates and anchorage-dependent growth in epithelial cells. Phosphorylates beta-1 and beta-3 integrin subunit on serine and threonine residues, but also AKT1 and GSK3B.

Tissue specificity

Highly expressed in heart followed by skeletal muscle, pancreas and kidney. Weakly expressed in placenta, lung and liver.

Immunocytochemistry/ Immunofluorescence - Integrin linked ILK antibody (ab2283)

ICC/IF image of ab2283 stained NIH/3T3 cells. The cells were methanol fixed (5 min) and incubated with the antibody (ab2283, 1µg/ml) for 1h at room temperature. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Image-iTTM FX Signal Enhancer was used as the primary blocking agent, 5% BSA (in TBS-T) was used for all other blocking steps. DAPI was used to stain the cell nuclei (blue). Alexa Fluor® 594 WGA was used to label plasma membranes (red).

ab2283 clearly stains the cytoplasm as expected for Integrin linked ILK. Some nuclear staining is also seen. We have no explanation for this nuclear staining and we would appreciate any customer feedback.

ab2283 staining Integrin linked ILK in mouse intestinal adenoma tissue section by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections). Tissue underwent formaldehyde fixation before heat mediated antigen retrieval and then blocking (5 minutes of peroxidase block and 10 minutes of protein block) for 15 minutes at 20°C was performed. The primary antibody was used diluted 1/2000 and incubated with sample for 45 minutes at 20°C. A HRP conjugated goat polyclonal to rabbit IgG was used undiluted as secondary antibody.
