Application Notes
Indirect ELISA 1/1000 - 1/10000.
Sandwich ELISA Use at an assay dependent dilution. Can be paired for Sandwich ELISA with Mouse monoclonal [GIF-10] to Interferon gamma (ab9805). PubMed: 9458493Used as the detection antibody in conjuction with recommended pair. sELISA can be used to determine IFN-g concentration in the cultures of human peripheral blood mononuclear cells.
Inhibition Assay Use at an assay dependent dilution. Can be used to neutralize the antiviral activity of IFN-g: The bioassay was performed using the L41 cell line and mouse encephalomyocarditis virus (EMCV). Briefly, interferon was pre-incubated with the ab for 2 h at RT and then added to the cell monolayer. 24 h later cells were infected with EMCV and cultured for further 24 h. Antiviral activity of interferon was evaluated using crystal-violet staining. PAbs to respective interferons and MAb isotype control were used as controls.
WB 1/500 - 1/1000. Detects a band of approximately 16-21 kDa.

Target

  • FunctionProduced by lymphocytes activated by specific antigens or mitogens. IFN-gamma, in addition to having antiviral activity, has important immunoregulatory functions. It is a potent activator of macrophages, it has antiproliferative effects on transformed cells and it can potentiate the antiviral and antitumor effects of the type I interferons.

  • Involvement in diseaseIn Caucasians, genetic variation in IFNG is associated with the risk of aplastic anemia (AA) [MIM:609135]. AA is a rare disease in which the reduction of the circulating blood cells results from damage to the stem cell pool in bone marrow. In most patients, the stem cell lesion is caused by an autoimmune attack. T-lymphocytes, activated by an endogenous or exogenous, and most often unknown antigenic stimulus, secrete cytokines, including IFN-gamma, which would in turn be able to suppress hematopoiesis.
