Recommended dilutions IP: Use at 2 - 5µg/mg of lysate. WB: 1/2000 - 1/10000. Detects a band of approximately 200 kDa (predicted molecular weight: 176 kDa).
Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Relevance
|
KDM5C / JARID1C / SMCX is expressed in a variety of tissues and it's highest levels are found in skeletal and brain extracts. Defects in SMCX are the cause of X-linked mental retardation (XLMR). Etiologically, mental retardation is a very heterogeneous condition that involves environmental, stochastic and/or genetic factors.
KDM5C specifically demethylates di and tri methyl histone H3 at K4.

Western blot - KDM5C / Jarid1C / SMCX antibody (ab72152)
All lanes : KDM5C / Jarid1C / SMCX antibody (ab72152) at 0.04 µg/ml
Lane 1 : HeLa whole cell lysate at 50 µg Lane 2 : HeLa whole cell lysate at 15 µg Lane 3 : HeLa whole cell lysate at 5 µg
Predicted band size : 176 kDa Observed band size : 200 kDa (why is the actual band size different from the predicted?) Additional bands at : 220 kDa,300 kDa. We are unsure as to the identity of these extra bands.

Immunoprecipitation - KDM5C / Jarid1C / SMCX antibody (ab72152)
Detection of Human KDM5C / Jarid1C / SMCX by Immunoprecipitation in Whole cell lysate from HeLa cells (1 mg/IP, 20% of IP loaded) using ab72152 at 3 µg/mg lysate for IP and at 0.1 µg/ml for subsequent WB detection. Note: bands between 55 and 71 kDa represent the IgG chains.
