Recommended dilutions ChIP: Use at an assay dependent dilution (PMID 17641388). IHC-P: Use at a concentration of 1 µg/ml. Perform heat mediated antigen retrieval via the pressure cooker method before commencing with IHC staining protocol (See image legend for further details). IHC-Fr: 1/100.
Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

KMT2H / ASH1 (absent, small, or homeotic 1) and Trithorax (trx) are members of the Drosophila trx group. Their products are associated with chromosomes and are believed to activate transcription of target genes through chromatin remodeling.
KMT2H / ASH1 is expressed in multiple tissues. The protein contains a SET domain, a PHD finger, four AT hooks and a region with homology to the bromodomain.

Immunohistochemistry (Frozen sections) - ASH1 antibody (ab4477)
Immunofluorescent staining for ASH1 on mouse neocortex (E12.5 whole brain frozen sections) using Rabbit polyclonal to ASH1 (ab4477). Frozen brain sections were post-fixed in paraformaldehyde; blocking was performed with 5% milk (30mins). ab4477 was used at a dilution of 1/100, incubated for 14h at 4°C. Secondary antibody was Alexa fluor 555 (1/500). Speckled expression of ASH1 was observed in the nuclei of cells throughout the developing CNS, neocortex pictured as reported in the literature [PMID: 10860993] Dean Griffiths, Org unknown, Country unknown

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - ASH1 antibody (ab4477)
IHC image of ab4477 stained human hippocampus FFPE section. Section was pre-treated using pressure cooker heat mediated antigen retrieval with sodium citrate buffer (pH6) for 30 seconds at 125°C. Section was incubated with ab4477, 1µg/ml, for 1h at room temperature and detected using an HRP conjugated polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
