Recommended dilutions
GSA: Use at an assay dependent dilution. Supershifts a specific methyl-CpG complex.
ICC: Use at a concentration of 1 µg/ml for positive nuclear staining in NIH3T3 cells fixed with 100% methanol.
IP: Use at an assay dependent dilution.
WB: Use at a concentration of 0.5 - 2 µg/ml on RIPA lysates from 3T3/A31 cells (ECL). Detects a band of approximately 97 kDa (predicted molecular weight: 75 kDa).

Not tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.

3T3/A31 cell lysate. Add 2.5µl of 2-mercaptoethanol/100µl of lysate and boil for 5 minutes to reduce the preparation. Load 20µg of reduced lysate per lane for minigels.

Relevance 

Kaiso is Caribbean slang for calypso music, which accompanied the late-night cloning of this gene. The deduced 672-amino acid human protein is 87% identical to the mouse protein, with mostly conservative changes. Kaiso contains an N-terminal POZ/BTB domain and 3 C-terminal zinc finger motifs. Immunoprecipitation analysis showed that Kaiso interacts with Ctnnd1 but not with Ctnnb1, Ctnna1 or Cdh1. Immunofluorescence analysis demonstrated a diffuse nuclear localization. Northern blot analysis of mouse tissues revealed ubiquitous expression of 5 to 6kb transcripts. Yeast 2 hybrid analysis indicated that the armadillo repeats of Ctnnd1 interact with a region flanking the zinc finger domain of Kaiso. By genomic sequence analysis, Daniel and Reynolds (1999) mapped the human KAISO gene to Xq23. They mapped the mouse Kaiso gene to the X chromosome also.

Kaiso is a component of the human N-CoR complex. In vitro, the Kaiso/N-CoR complex binds specific CpG-rich sequences in a methylation-dependent manner. In vivo, Kaiso targets the N-CoR complex to the MTA2 gene promoter in a methylation-dependent manner. Kaiso is required for transcriptional repression of the methylated MTA2 locus. Furthermore, this repression also requires a functional N-CoR deacetylase complex, which brings about histone hypoacetylation and methylation of H3 lysine 9 to the MTA2 locus (Yoon HG et al. 2003).
