  • Application notesWB: 1/1000 when using colorimetric substrates such as BCIP/NBT and 1/5000 for chemiluminescent substrates. Predicted molecular weight: 29 kDa. Dilution optimised using Chromogenic detection. A range of different sized bands can be seen for Kallikrein 1 in different tissues and in cell culture media, ranging from 16-38 kDa and likely representing different proteolytically processed forms. SDS-PAGE: Use at an assay dependent dilution. Extensive glycosylation and other posttranslational modifications give Kallikrein an apparent mass of 36-46 kDa on SDS PAGE gels. Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

    • FunctionGlandular kallikreins cleave Met-Lys and Arg-Ser bonds in kininogen to release Lys-bradykinin.
    • Tissue specificityIsoform 2 is expressed in pancreas, salivary glands, kidney, colon, prostate gland, testis, spleen and the colon adenocarcinoma cell line T84.
    • Sequence similaritiesBelongs to the peptidase S1 family. Kallikrein subfamily.

      The O-linked polysaccharides on Ser-93, Ser-104 and Ser-167 are probably the mucin type linked to GalNAc. In PubMed:3163150, GalNAc was detected with the corresponding peptides but not located.
