Are these antibodies likely to cross-react with mouse proteins?

Following up my previous message, immunogens for the kallikrein antibodies ab28289 and ab40497 were aligned with the mouse canonical sequence (http://www.uniprot.org/uniprot/P15947.fasta).

The ab28289 epitope falls in a region of relatively low sequence homology, approximately 48 % identical, so the antibody is unlikely to recognize the mouse kallikrein very well, if at all. The ab40947 immunogen homology is better, with 66% identity, and may recognize the mouse protein, although not as well as the human protein.

Are these antibodies likely to cross-react with mouse proteins?

I am still waiting for our source for the kallikrein antibodies ab28289 and ab40497 to send their alignments of the immunogens and mouse kallikrein but I have checked the homology of the immunogens for the other four antibodies against the corresponding mouse sequences.

For the third kallikrein antibody we discussed, ab59520, the homology is relatively low, with only 58% identity:

PN++C KAH++K

The letters are the amino acids that are conserved and the "+" symbols are biochemically similar amino acids, but not identical. There is some chance of cross-reaction but we feel it is unlikely.

For the remaining three antibodies, ab103013 anti-PTPRD, ab1100844 anti-troponin C, and ab116610 anti-ITIH4, the alignment results are as follows:

ab103013: 93% identity ab110844: 99% identity ab116610: 50% identity.

The first two are likely to react with the mouse proteins, but we do not guarantee this.
