Our Abpromise guarantee covers the use of ab66620 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
ICC/IF: Use a concentration of 5 µg/ml
WB: Use a concentration of 1.0 µg/mlDetects a band of approximately 70 kDa (predicted molecular weight: 70 kDa).
IHC-P: Use a concentration of 4 - 8 µg/ml.
Retains NFE2L2/NRF2 in the cytosol. Functions as substrate adapter protein for the E3 ubiquitin ligase complex formed by CUL3 and RBX1. Targets NFE2L2/NRF2 for ubiquitination and degradation by the proteasome, thus resulting in the suppression of its transcriptional activity and the repression of antioxidant response element-mediated detoxifying enzyme gene expression. May also retain BPTF in the cytosol. Targets PGAM5 for ubiquitination and degradation by the proteasome.
Broadly expressed, with highest levels in skeletal muscle.
Contains 1 BACK (BTB/Kelch associated) domain.
Contains 1 BTB (POZ) domain.
Contains 6 Kelch repeats.
The Kelch repeats mediate interaction with NF2L2/NRF2, BPTF and PGAM5.
Ubiquitinated and subject to proteasomal degradation.
Cytoplasm. Nucleus. Shuttles between cytoplasm and nucleus.

Immunocytochemistry/ Immunofluorescence-Keap1 antibody(ab66620)

ICC/IF image of ab66620 stained HepG2 cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab66620, 5µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
