Question 1

Wednesday 02-May-2012

Looking for an antibody against the extracellular epitope of LAMP1, to work in flow cyt, on mouse and human tissue.

ANSWER:

Thank you for contacting Abcam.

I have heard back from the about whether some of our LAMP1 antibodies recognize an extracellular epitope or not.

Although for ab25630 & ab25245, we have not performed epitope mapping; we have determined through QC testing that ab25630 recognizes an extracellular epitope on both human activated platelets and U937 cells (which adopt the morphology and characteristics of mature macrophages). We also know ab25245 to recognize an extracellular epitope on mouse epithelial cells and macrophages.

Please let us know if you have any other questions. I am still waiting to hear back about ab119127, and I will let you know as soon as I know more. Also, attached is the Abcam protocols book, that we discussed.

Question 2

Monday 30-January-2012

I am currently using the ab25245, Rat anti-lamp1/CD107A antibody for western. And now I am confused which is the right band size.
On the main page of antibody information, it is decribed that the expected band size is 40kDa, but in your image info, the band size looks around 100kDa.
And another antibody such as ab24170 shows the band size should be around 120kDa.
It looks the band size really depends on which antibody I am using, but

please let me know which one is the right size in case of ab25245. Thanks

ANSWER:

Thank you for contacting Abcam regarding ab25245.

I can confirm that according to SwissProt, the predicted molecular weight of LAMP1 is indeed ˜40kDa. However, this protein is subject to glycosylation at numerous site which accounts for the large shift in MW. We would therefore expect a band running at approximately 110 - 140kDa, consistent with ab24170.

I hope this information is helpful and clarifies any confusion. Please do not hesitate to contact us if you have any additional questions or concerns.

Question 3

Friday 08-June-2007

i ordered the anti lamp-1 antibody ab25245 recognizing the mouse lamp-1. U cite the pubmed reference 17215451, but they said they used an anti-human lamp-1 antibody from abcam for their human Hela cells, so i don't think that is this one? Or they did a mistake? Could u let me know when u find out please?

ANSWER:

Thank you for your enquiry.

Due to my limited access to the publication, I was not able to confirm if the authors indeed used our product with human Hela cells. It could be that they tested our product with human samples (if they quoted our catalogue number) and managed to obtain results but we did not add that new information on our datasheet. Or, they could have quoted our product code wrongly.

In any case, I have asked our marketing department to look into it. If we have left out any new information, we will include that in the datasheet. On the other hand, if the authors have made an error, the reference will be removed from our datasheet.

Please rest assure that this product has been tested and we guarantee that it will work in mouse species. If you have any other questions, please do not hesitate to contact me.
