Recommended dilutions
ICC: 1/100. Fix cells with methanol or with paraformaldehyde-lysine-periodate formulation. If the latter is used, permeabilize with saponin, not with Triton X-100 or NP-40. The antibody will label the presumptive lysosomes and late endosomes in cells that have been permeabilized with saponin 0.01%.
IHC-FoFr: 1/100 (PubMed:17425404).
IP: Use at an assay dependent dilution (PMID 2142158).
WB: Use at an assay dependent dilution (see Abreview).


Not yet tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.

Relevance 

Lysosomes are membrane bound vacuoles which play a critical role in cellular metabolism. These organelles participate in endocytosis, the synthesis and assembly of acid hydrolases, act as sites for digestion of foreign materials and for specialized autolytic cellular processes. Membrane-associated glycoproteins have an important role in normal lysosomal function. Prior to post-translational modification, Lysosome Associated Membrane Protein 2 (LAMP2) is a ~45 kDa polypeptide. Mature, functional LAMP2 is extensively glycosylated with a variety of different N linked and O linked oligosaccharides with a total molecular weight of ~100-110 kDa. LAMP2 has a structure comprised of a large amino terminal intra lysosomal domain, a hydrophobic transmembrane domain, and a short carboxyl terminal cytoplasmic tail. Studies suggest that LAMP2 may be involved in leucocyte adhesion.
LAMP2 has recently been shown to be important in autophagy - the process of bulk degradation of cellular proteins. Human LAMP-2 deficiency causes Danon’s disease which is associated with the accumulation of autophagic
material in striated myocytes.
