This product has not worked for me. There are no bands in my blot. I am confident of good transfer and loading. The product also is brown. Could you give me advice or a new antibody?

Where did you see this protein appear on Western blots using this antibody?

ANSWER:

 

Thank you for your recent technical service inquiry.

According to data from our source this product will react with lactate dehydrogenase from rabbit muscle showing a 138kDa band under non-reducing conditions and a 36kDa band under reducing conditions. Although they do not have a picture available, information from a personal communication shows a 36kDa band when this product was used at 1:1000 in HeLa cell extracts. The reactivity was observed in the cytoplasmic fraction, and not in the nuclear fraction. About 30ug of lysate was loaded per lane onto a 10% gel for SDS-PAGE.

It is possible that the 55kDa band you are seeing is LDH. My recommendation would be to perform a peptide competition assay to see if the band competes out after pre-incubating with the protein. Biocompare has links to several companies that provide multiple kinds of lactate dehydrogenase:

We just ordered two primary antibodies from Abcam, ab7671 and ab7639. I was wondering whether abcam has the original proteins, Sodium-Potassium ATPase and Lactate Dehydrogenase, that inteact with those two antibodies. I appreciate if you could assist us with information about where we can get Sodium-Potassium ATPase and Lactate Dehydrogenase, as we need controls when using the primary probing. I guess the best way to find out the controls is to ask Abcam about any availible information.
