Cytoplasm. Nucleus. Predominantly cytoplasmic at G1 phase, accumulates in the nucleus in S and G2 phases. The frequency of nuclear localization in S and in G2 phases is 60% and 30%, respectively.

Relevance 

Lin28B acts as a suppressor of microRNA (miRNA) biogenesis. Lin28B binds to the miRNA precursor pre-let-7 and recruits the uridylyltransferase ZCCHC11/TUT4, resulting in the terminal uridylation of pre-let-7. Uridylated pre-let-7 miRNAs are not processed by Dicer and are subsequently degraded. Lin28B recognises the 5'-GGAG-3' motif present in the terminal loop of pre-let-7. It also recognizes, and binds to, other pre-miRNAs that contain this motif in the terminal loop, leading to their terminal uridylation and subsequent degradation. Lin28B mediates Myc-mediated let-7 repression. Overexpression of Lin28B has been observed in many cancers, and is linked to repression of the let-7 family of miRNAs and derepression of let-7 targets.
