I have a customer looking for an alkaline phosphatase conjugated lysozyme antibody, and they specifically asked about AB391. I don't see any alk phos conjugates listed on your web, but they have also asked if you can custom conjugate this antibody if a conjugate is not available. Can you please let me know asap? Yes, this antibody has been tested by immunoprecipitation and Western blot. The suggested working dilution is 1/4000 to 1/10000, although optimisation in your own lab is recommended. |

I wanted to ask about the anti-Lysozyme (product code:ab391, batch 5732) which I recently purchased from you. It appears by Western blotting that although the antibody reacts strongly with egg white lysozyme (detects ca. 0.1ng), there is also reaction with many E. coli proteins. I wanted to ask whether this result could be due to bacterial material in the adjuvant used in immunization? Or is this result unexpected or incorrect? I am using this antibody in conjunction with cloning in E. coli, so that the cross-reaction is a definite problem. Thank you for any information or assistance you can provide. |

Polyclonal antibodies are normally produced in SPF (specific pathogen free) rabbits and these rabbits are free of infection with E. coli. Even with these precautions, quite a lot of polyclonal antibodies give cross-reaction with E. coli proteins. Try using less of the antibody and/or shorter incubation times to reduce the background. If this doesn't help, a good method is to adsorb the antibody with a E. coli protein preparation that does not contain your antigen of interest. |

This antibody is known to work in immunoblotting, however it's reactivity against dog lysozyme has not been determined. As the antibody was raised against lysozyme from chicken egg white, it is uncertain whether it will react with canine. |
