Question 1

Friday 06-July-2007

Do you have the epitope information for this product?

ANSWER:

Thank you for your enquiry.

To our knowledge the epitope which is recognized by Rl-ph1 is not known. The antibody response which led to the induction of Rl-ph1 was elicited against a total phage M13 extract and not to a specific epitope of the g8p protein and as far as I know no articles exist in which epitope mapping was done.

Question 2

Wednesday 07-September-2005

BATCH NUMBER -- NOT SPECIFIED -- ORDER NUMBER -- NOT SPECIFIED --

DESCRIPTION OF THE PROBLEM No signal

SAMPLE M13 phage

PRIMARY ANTIBODY ab9223, 1:1000 in blocking buffer, RT, 1hr

DETECTION METHOD ECL

ANTIBODY STORAGE CONDITIONS -20C

SAMPLE PREPARATION phage solution in PBS

AMOUNT OF PROTEIN LOADED >15 mg

ELECTROPHORESIS/GEL CONDITIONS dot blot analysis, not required

TRANSFER AND BLOCKING CONDITIONS dot blot analysis, not required 15% skim milk in PBS

SECONDARY ANTIBODY Sigma, Goat anti-mouse IgG HRP conjugate, 1:1000 in blocking buffer , RT, 1hr

HOW MANY TIMES HAVE YOU TRIED THE APPLICATION? 3 HAVE YOU RUN A "NO PRIMARY" CONTROL? No DO YOU OBTAIN THE SAME RESULTS EVERY TIME? Yes

WHAT STEPS HAVE YOU ALTERED? none

ADDITIONAL NOTES Customer also spotted ab9223 directly on the membrane and stained with HRP anti mouse 2nd antibody but there was no signal either.

The secondary antibody works fine with other mouse primary antibodies.

ANSWER:

Thank you for your enquiry. Is your customer using this antibody in Western blotting or in dot blot? The antibody has only so far been tested for application in Western blotting and Immunohistochemistry (Frozen sections).

I do need some additional details as well. What was the lot number? For the amount of protein loaded. does your customer mean 15 ug? How was the sample prepared? Has your customer tried extending the antibody concentrations and incubations periods?

Question 3

Tuesday 19-March-2002

Dear M.,

I would like to have some informations about three of your products: anti-M13 Filamentous Phages coat protein pVIII purified ab9223, ab9224 and ab 9225. Could you give me the concentration used or an ELISA and a Western Blot. I also would like to know wich part of the pVIII is binded by these Mab.

Thank you in advance, Thierry Laurent

ANSWER:

In all three cases, all the information we have is on the datasheet. The epitope for each of these antibodies has not been mapped and all are supplied as tissue culture supernatents. We recommend the researcher to optimise conditions for ELISA and WB.
