Recommended dilutions
ELISA: Use at a concentration of 0.1 - 1 µg/ml.
ICC: 1/20.
ICC/IF: Used at a dilution of 1/25 (see Abreview).
IHC-P: 1/100. Perform heat mediated antigen retrieval before commencing with IHC staining protocol.
IP: Use 3µg for 107 cells.
WB: Use at a concentration of 0.5 - 2 µg/ml. Predicted molecular weight: 34 kDa.


Not yet tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.

Relevance 

The MAGE 1 gene is a member of the melanoma antigen encoding gene family.
These genes encode for HLA-restricted tumor associated rejection antigens recognized by cytotoxic T lymphocytes. Some of these target antigens may be potentially useful for cancer specific immunotherapy. The expression of MAGE genes has been reported not only in melanoma but also in various other malignant tumors such as hepatocellular carcinoma and germ cell tumors.

ab21472 staining the human breast cancer cell line, MCF-7, by ICC/IF. The cell line was stimulated with 5-aza-CdR for 72 hours. The treated cells, or a mock control were then stimulated by trichostatin A for 24 hours. ab21472 was used at a dilution of 1/25 and an Alexa Fluor ® 546 conjugated goat anti-rabbit IgG was used as the secondary.
