Specificity 

This antibody is highly specific to MAP 2 a + b and shows no cross-reaction with MAP 1, MAP 5, tubulin, or tau.
It reacts with dendrites and cell bodies of neurons but not with the neuronal processes.
It does not react with the low molecular weight (70kDa) form (2c) of MAP 2.

Recommended dilutions
ICC/IF: Use at an assay dependent dilution (see review).
IHC-P: Use at a concentration of 1 - 2 µg/ml (30mins at RT). Perform heat mediated antigen retrieval before commencing with IHC staining protocol (10mM citrate buffer pH 6.0, for 10-20 min followed by cooling at RT for 20 mins).
IHC-Fr: 1/200.
WB: Use at a concentration of 1 - 2 µg/ml ( 2 hours at RT). Detects a band of approximately 280 kDa.

Not tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.

Relevance 

MAP2 is the major microtubule associated protein of brain tissue. There are three forms of MAP2; two are similarily sized with apparent molecular weights of 280 kDa (MAP2a and MAP2b) and the third with a lower molecular weight of 70 kDa (MAP2c). The expression of MAP2 is developmentally regulated and its multiple forms arise by alternative splicing of a single gene.

This image was taken with a 5x objective and is from the CA3 region of rat hippocampus. It is possible to see cell soma slightly stained and the dendrites are also well stained. Only pyramidal cells are labelled here and there are no processes from interneurons. This is not a confocal reconstruction.

ab3096 at a 1/200 dilution staining MAP2a + MAP2b in whole rat embryo tissue sections by Immunohistochemistry (frozen sections) incubated at 25°C for 16 hours. PFA fixed. Blocked using 5% serum for 1 hour at 25°C. Secondary used at 1/200 polyclonal Goat anti-mouse conjugated to Alexa Fluor 546.
