Target

    Exposure time : 2 minutes

    This blot was produced using a 4-12% Bis-tris gel under the MOPS buffer system. The gel was run at 200V for 50 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 2% Bovine Serum Albumin before being incubated with ab22354 overnight at 4°C. Antibody binding was detected using an anti-rabbit antibody conjugated to HRP, and visualised using ECL development solution ab133406.

  • ab22354 staining MASPIN in human colon tissue by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded tissue sections). The sections were fixed in paraformaldehyde and subjected to heat-mediated antigen retrieval in TRIS-EDTA Buffer pH 9.0 and permeabilized with a wash buffer with Tween. The primary antibody was diluted 1/2000 and incubated with the sample for 30 minutes at 20°C. An HRP-conjugated Goat anti-Rabbit polyclonal was used as the secondary antibody.

    Observed band size : 40 kDa (why is the actual band size different from the predicted?)
    Additional bands at : 30 kDa (possible cleavage fragment).
    ab22354 recognizes a band at approximately 40 kDa that corresponds in size to MASPIN (molecular weight of 42 kDa). This band is competed away by the addition of the immunizing peptide suggesting that the interaction is specific.
