We purchased a Madcam1 antibody ab34143 [17F5] from Abcam a few weeks ago. I’m very sorry to tell you this, but this antibody is not so working well.. It is actually worse than another madcam1 antibody ab34247 [314G8] which we bought last year. With the old madcam1 antibody (314G8), we saw at least low levels of Madcam1 signal on some stromal cells and endothelial cells, but the new madcam1 (ab34143) is just not showing anything at all.

I checked the Abcam’s website and saw that ab34143 has a reference in the following paper:

Allavena R et al. CNS elevation of vascular and not mucosal addressin cell adhesion molecules in patients with multiple sclerosis. Am J Pathol 176:556-62 (2010). IHC-Fr; Human.Read more (PubMed: 20035048)

However, this paper only shows the stainings with ab34247 [314G8], but not with ab34143 [17F5]. Maybe because the performance of ab34143 [17F5] is not as good as the other one? Could you please check with Abcam if 17F5 is validated for IHC-fr and help me to solve this issue?

I am sorry to hear you are experiencing difficulties with ab34143 in IHC-Fr. This antibody is garanteed to work on Human samplesin IHC-Fr, Flow Cyt, ELISA, IHC-P andWB.
