Question 1

Monday 28-February-2005

I have a question about two antibodies against MBD1 you offer. The catalog number for the first one is ab3753 and the other is ab2846 both detect the same protein with all his isoforms, but the molecular weight is very different. The first one ab3753 shows a band at 60kDa [your website says: Synthetic peptide: KQEPPDPEEDKEENKDDSAS (Human). This represents the C-terminal amino acids of all human MBD 1 isoforms. WB: Use at a dilution of 1/1000. Detects a band of approximately 60 kDa (predicted molecular weight: 57.6 kDa)]. and the other ab2846 at 83kDa [your website: Synthetic peptide: C-RSKDLKKPGARKQ, corresponding to amino acids 593-605 of Human MBD 1 WB: Use at a concentration of 2 - 5 ?g/ml. Detects a band of approximately 83 kDa (predicted molecular weight: 83 kDa)]. I hope you are able to explain this.

ANSWER:

Thank you for your enquiry and for your patience. I have been looking into the issue that you have raised. Both ab3753 and ab2846 are originated outside Abcam and by two different sources.

The originator of ab2846 said that all evidence that they have shows this antibody detecting a band at ~83 kDa. The SwissProt database link does say the molecular weight is 57.6 kDa. Ab2846 was only tested on recombinant MBD1 protein which may be the reason for the different molecular weights in the antibodies.

I'm still investigating this issue and will let you of any additional information that I obtain.
