The western blot analysis of alkaline phosphatase treated extracts show clearly that this antibody specifically detects phosphorylated MCM2. However, can you confirm that this antibody differentially detects phospho MCM2 during cell cycle (in G1/S MCM2 should be under phosphorylated but in S phase it is hyperphosphorylated).

This antibody has been tested and shown to work by western blotting only. The specificity of this antibody was tested using western blot extracts. The specificity of this antibody for S phase cells on account of MCM2 phosphorylation has not been determined.

Customer is using these two antibodies in Western blotting with HCT16 lysate. For both she is seeing a doublet at 125 kDa.

Thank you very much for your enquiry. The originator of these two antibodies has provided me with the following information. To their knowledge they have not heard of reports of doublets being seen with ab11897 and ab11896. Both of these were tested using Hela cell lysates and from these results, one might perceive a doublet but the resolution is insufficient to make a clear judgement.

This is all the information that I have at the moment. Have you tried these two antibodies with different types of samples?
