Dr. Miller- I have since run controls on this antibody, and it appears to be staining all tissues the same. The application I am using it for is isolated islets, but the same staining patterns appear (very bright too) for normal kidney tissue. Since I know the islets should be positive, I can only conclude the antibody is not working for fluorescence IHC. I've had quite a few other people specializing only in fluorescence IHC, and they have came to the same conclusion. |

In addition to the information I sent through to you yesterday regarding two alternative antibodies that you may be interested in, I have now received more information from the originator regarding the use of ab7814 in IHC: The following articles feature the use of this antibody to detect MCP-1 on frozen sections: - Expression of Monocyte chemoattractant protein-1 in delayed type hypersensitivity reactions in the skin. Xiaohui Yu et al.; Laboratory investigation, 1992. Vol 71, No. 2, p226. - A new monoclonal antibody (5D3-F7) which recognizes human monocyte-chemotactic protein-1 but not related chemokines. Development of a sandwich Elisa and in situ detection of producing cells. Giuseppe Peri et al. |

I have chased the originator for further information about how ab7814 was used in IHC and will get back to you as soon as I receive their reply. There are two other antibodies against MCP-1 that may be useful for your research: ab7202 -- Rabbit polyclonal to rat MCP 1 / MCAF ab 8102 -- Mouse monoclonal (MNA1) to human MCP 1 / MCAF Please note ab7202 has only been tested for cross reactivity with rat and mouse. Both antibodies are recommended for use in IHC on frozen sections. Please let me know if you would like a complementary vial of either of these antibodies. |

Dear sirs, is it possible to use the rabbit polyclonal anti-human MCP-1 antibody (ab7814) for formalin-fixed paraffin-embedded sections (immunohistochemistry)? Thank you for your answer.
|
|||||||||
ANSWER: |
This antibody has not been tested on formalin fixed paraffin embedded tissues, therefore we can't guarantee results.

Dear sir: Thank you so much for your kindly help, but what I want is the antibody to rabbit MCP-1 because I have used the rabbit as the animal athrosclerosis model and I have to detect the MCP-1 in these rabbit aortas, so, would you please check whether there are mono- or poly-clone antibodies to rabbit MCP-1 or cross-reactive antibodies to rabbit? (ab7814 is the polyclone antibody to human MCP-1 and Cross-reactivity to MCP-1 of other species has not been determined) I look forward to your reply! Thank you very much! |

ANSWER: |
We have two antibodies to MCP-1: this one which has only been tested on human tissue, and ab7202 which is reactive to both rat and mouse (although again not tested on rabbits). We cannot guarantee that either of these will work on rabbit. |
