General notes

Stimulated macrophages produce MMP-12; it has also been found in osteosarcoma cells, synovial fibroblasts and lung fibroblasts. MMP-12 levels in quiescent cells and tissues are minimal (with the exception of macrophages), and mitogen stimulation or protein concentration is often needed to visualize the bands. In addition, cell types differ greatly in the quantity of MMP-12 produced.

Application notes

WB: Recommended starting dilution of 1/1000 (when using colorimetric substrates such as BCIP/NBT) and 1/5000 (for chemiluminescent substrates). Higher concentrations of antibody may be needed for non human samples. When used against the reduced protein, the antibody identifies bands at 54 kDa, 45 kDa, and activated forms in between (predicted molecular weight: 54 kDa). Dilution optimised using Chromogenic detection. Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Function

May be involved in tissue injury and remodeling. Has significant elastolytic activity. Can accept large and small amino acids at the P1' site, but has a preference for leucine. Aromatic or hydrophobic residues are preferred at the P1 site, with small hydrophobic residues (preferably alanine) occupying P3.

Tissue specificity

Found in alveolar macrophages but not in peripheral blood monocytes.

Belongs to the peptidase M10A family.
Contains 4 hemopexin-like domains.

Domain

The conserved cysteine present in the cysteine-switch motif binds the catalytic zinc ion, thus inhibiting the enzyme. The dissociation of the cysteine from the zinc ion upon the activation-peptide release activates the enzyme.
