Can you tell me the antigen retrieval method used?

The antigen retrieval method used for this product was pressure cooker method. Please find the details of the protocol below.

Pressure cooker antigen retrieval protocol.

Solution: Tris-EDTA Buffer (10mM Tris Base, 1mM EDTA Solution, 0.05% Tween 20, pH 9.0)

Tris 1.21 g

EDTA 0.37 g

Distilled water 1000 ml (100 ml to make 10x, 50 ml to make 20x)

Mix to dissolve. pH is usually at 9.0. Add 0.5 ml of Tween 20 and mix well. Store at room temperature for 3 months or at 4C for longer storage.

Protocol:

1. Immerse slides into staining dish containing Antigen Retrieval Solution.

2. Place covered staining dish into the rice cooker. Add 120 mL dH2O and press “cook”.

3. When “cook” is turned to “warm” (about 20–30 min), unplug the cooker and remove the staining dish to the bench top.

4. Allow to cool down, without cover, for 20 min.
