WB: A recommended starting concentration for Western blots is 1:2,000 when using colorimetric substrates such as BCIP/NBT, and 1:5000 for chemiluminescent substrates. Higher concentration of antibody may be needed for non-human samples Identifies bands at 65 Kd. Dilution optimised using Chromogenic detection. Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Seems to specifically activate progelatinase A. May thus trigger invasion by tumor cells by activating progelatinase A on the tumor cell surface.

Expressed in stromal cells of colon, breast, and head and neck.

Domain

The conserved cysteine present in the cysteine-switch motif binds the catalytic zinc ion, thus inhibiting the enzyme. The dissociation of the cysteine from the zinc ion upon the activation-peptide release activates the enzyme.

The precursor is cleaved by a furin endopeptidase.

Membrane. Melanosome. Identified by mass spectrometry in melanosome fractions from stage I to stage IV.

ab38969 staining Propeptide domain in human fibrosarcoma cells by Immunocytochemistry/ Immunofluorescence. Cells were fixed with paraformaldehyde, permeabilized with 0.1% Triton x100 and no blocking was done. Samples were incubated with primary antibody diluted to 10µg/ml for 1 hour at 20°C. An Alexa Fluor®555-conjugated donkey polyclonal to goat IgG was used undiluted as secondary antibody.
